Chronic infections with the hepatitis B virus (HBV) affect 250 million people worldwide, making it one of the most prevalent viral pathogens in men. These chronic infections are the major cause of liver cirrhosis and hepatocellular carcinoma. HBV is a non-cytopathic virus and HBV-related liver damage is caused by immune-mediated events during clearance of infected hepatocytes, and a role for virus-specific T cells has been firmly established. However, the involvement of B cells in controlling the HBV infection and in immunopathology of the liver is still elusive.
The aim of this study is to obtain insight into the role of B cells in HBV patients, through molecular characterization by immunological techniques, single cell RNASeq and B cell receptor sequencing of blood and liver virus-specific B cells of patients infected with HBV, and multiplex immunostainings. This project will lead to a better understanding of the role of B cells during chronic infection, which may guide improved treatment strategies aimed at therapeutic vaccines inducing antibody responses.
The research combines translational immunological studies on material from a large outpatient viral hepatitis cohort with clinical trials at the Erasmus MC in which novel antiviral compounds and treatment strategies are being evaluated.
Work environment
The laboratory of Gastroenterology and Hepatology of the Erasmus MC is a unique environment for high-level biomedical research on liver and gastrointestinal disease.
